8页1、编号:时间:2021年x月x日书山有路勤为径,学海无涯苦作舟页码:第1页 共1页生物转化法合成谷胱甘肽的研究 张磊 薛钊坤 (浙江工业大学生物与环境工程学院 杭州 310014)摘要:本文以编号X23的藤黄八叠球菌为研究对象,研究其生物转化合成谷胱甘肽的可能性。通过从物理、化学和生物三方面因素改进转化条件:转化过程中以100mg/L湿菌含量加入新配制pH为7.5的转化液137、转速200r/min的摇床转化6h,用超声波对含菌的转化液破壁10min,接着于3500r/min离心机离心20min后取上清液,为最佳的物理因素;通过正交实验分析方法得转化液最佳成份浓度为葡萄糖0.75mol/L、MgCl20.01mol/L、底物浓度0.03mol/L,找到并证实微量元素浓度20mg/L的ZnSO47H2O对谷胱甘肽合成酶有激活作用,为最佳化学因素;连续生物转化进行两步可使转化产量达到88.43mg/L,一次从发酵到转化两步的生产过程可产谷胱甘肽231.27mg/L,较原菌株只发酵的产量142.75mg/L提高61.94%。关键词:谷胱甘肽、藤黄球菌、生物转化RESERCH OF BIOLOG

3、iquid pH 7.5 contained Sarcina lutea 100mg cell/L,was placed in a flask and incubated at 37with shaking at 200r/min for 6h,then the strain with productive liquid was smashed for 10min by sonicate, supernatent was got by centrifugation at the speed of 3500r/min for 20min,that was the optimum physical factor. Composition of the productive liquid, Glucose 0.25mg/L、MgCl2 0.01mg/L、Amino acids 0.03mg/L. was obtained by the quadratic design. the ZnSO47H2O at the concentration of 20mg/L which was discov
4、ered and confirmed can activate the endosynthase of glutathione, that was the optimum chemical factor. The result indicated that the glutathione production reached 88.43 mg/L after two steps biologic transform process, The productivity of glutathione can be obtained 231.27 mg/L of during the whole process, which was 61.94 higher than in the synthesis.
