2页豬瘟抗體 ELISA 之檢測應用豬瘟研究組 蕭雅如 約聘研究助理摘 要為加強豬場豬瘟防疫,我國以 LPC 疫苗免疫來防範豬瘟的發生,而豬瘟抗體的監測是以血清中和試驗為主,但由於血清中和抗體檢測法費時費力,因此本研究之目的,藉由比較市售 IDEXX®豬瘟抗體 ELISA 檢測套組與中和抗體間之相關性,進而評估市售豬瘟抗體ELISA 檢測套組應用於血清監控之可行性利用評估仔豬免疫 LPC疫苗後,來比較其血清中和抗體力價與豬瘟抗體 ELISA 檢測套組之相關性結果顯示 ELISA 抗體與中和抗體力價極為相似,檢測之結果相關係數 r 值為 0.827,以上結果顯示豬瘟抗體檢測在 ELISA 與中和抗體檢測間具有高度相關性利用 ELISA 檢測套組分析 2006年豬瘟送檢樣品 15 個豬場中,呈現免疫反應正常豬場 (陽性率≧80%)之上市肉豬血清 ELISA 抗體力價,抗體平均力價為 70.68%,可作為正常上市肉豬經免疫後抗體力價之參考監控潛在豬瘟污染場中高豬瘟抗體力價豬隻之實驗中,結果亦顯示血清經 80 倍稀釋後再以 ELISA 套組檢測,可有效區分高 ELISA 抗體力價≧80%之 LPC 疫苗正常免疫豬隻 (33.97 ±3.18%) 及豬瘟病毒感染耐過豬隻之血清 (60.23 ±4.77%),因此血清樣品經 80 倍釋稀後,若 ELISA 抗體力價≧47.02%時,則可懷疑曾遭受豬瘟病毒之潛在感染。
以上結果顯示豬瘟抗體檢測在 ELISA 與中和抗體檢測間具有相關性,可應用於豬瘟血清抗體監測、豬場免疫評估及遭受野外病毒污染之豬場監控上The application of ELISA test in CSF antibodyHog cholera division Ya Ju HsiaoPrevention with LPC vaccine and immunosurveillance for classical swine fever (CSF) are the most important issues in control and CSF eradication program in Taiwan. Serum neutralization (SN) test has been thought as the golden standard method for detection CSF, but it is not suitable for large-scale screening due to arduous and time-consuming. The experiment was aimed to compare the correlation between SN and commercial CSF antibody enzyme-linked immunosorbent assay (ELISA) kits and further evaluate the application of ELISA test on LPC vaccination program and other CSF immuno- surveillance. First, we compared the correlation between different levels of SN titers and IDEXX® CSF antibody ELISA titers. The results show that the correlation (r=0.827 ) between SN titers and IDEXX® ELISA kits,have a high r value, it is applied in following tests.We used ELISA kits to analysis serum sample from 52 farms in 2006. The result was showed that the mean of titer was 70.68% in 827 samples (collection from the farm whose >85% positive prevalence). This ELISA titer of marking pig vaccinated by LPC vaccine could be a reference data. In the other experiment, the ELISA tests were performed in CSF virus contamination farms. The results indicated that the IDEXX ELISA kit could differentiate the LPC- vaccinated (34.67±3.18%) from CSF virus experimentally infected pigs (58.62±4.77%). According to this result, the ELISA titer was more than 47.5%, it could be suspected CFS infection pig. These data was demonstrated that it was high correction between ELISA and SN test. It could be perform in large scale immunosurveillance, evaluation of LPC vaccination program and efficacy, and monitor the farm whether was persistent infection by CSF virus. It can be applied in large scale of immunosurveillance, evaluation of LPC vaccination program and efficacy, and, perhaps screen of CSF virus persistent infection in pig farms.。
