39页南昌大学医学院硕士学位论文抗氧化剂对NO缺乏性大鼠心肌氧化应激及p22phox表达的影响姓名:周斯平申请学位级别:硕士专业:内科学指导教师:吴延庆20080501摘要目的观察抗氧化剂依布硒咻(Ebselen, Ebs)、维生素E(VitE)对N-硝基7-精 氨酸甲酯(L-NAME)所致的一氧化氮(NO)缺乏性高血压大鼠心脏氧化应激及 NADPH (还原型辅酶II)氧化酶P22亚单位(p22phox)蛋白表达的影响方法Wistar大鼠40只,随机分为5组(n=8),正常对照组(C).实验对照组(L)、 依布硒咻组(E)、VitE组(V)两者联合预防组(D)组,灌胃法给药隔周称重 并用尾袖法测动脉收缩压8周末处死大鼠采集血标本,取心尖部心肌组织做匀 浆,测心脏匀浆液中的N0、血管紧张素II (AngHX丙二醛(MDA)、心肌组织 超氧阴离子(0T)的含量或活性,蛋白印迹法(Western blotting)测定心肌 组织中还原型辅酶II氧化酶p22亚单位(p22phox)蛋白表达测心脏重量/体 重(mg/g)指数结果①各实验组大鼠血压逐渐升高,2周末达到142.17・69 mmHg,但各组间无 差异;8周末比C组明显增加(P < 0. 01),抗氧化剂组比L组血压低(P < 0. 05), 联合抗氧化剂组血压显著低于单用组(P<0.01), Ebs组血压比VitE组低(P <0. 05)o②各实验组心肌组织NO水平明显低于C组(P<0.01),抗氧化剂组 明显高于L组(P<0・05),联合抗氧化剂组高于单一抗氧化剂组(P<0・05), Ebs组比VitE组高(P<0. 05)o③各实验组心肌组织AngII. MDA以及心肌组织 02-含量均显著高于C组(P<0. 05),抗氧化剂组低与L组(P<0.05),联合抗 氧化剂组低于单一抗氧化剂组(P < 0.05), Ebs组低于VitE组(P<0. 05)o④ 各实验组心脏重量/体璽指数显著高于C组(P<0.01),抗氧化剂组明显低于L 组(P<0.05),联合抗氧化剂组低于Ebs组(P<0.05), Ebs组低于VitE组(P <0. 05)o⑤心肌组织p22phox蛋白表达L组明显高于C组(P<0.05),抗氧化 剂组明显低于L组血压(P < 0.05),联合抗氧化剂组显著低于单用组(P < 0. 01),Ebs组血压比VitE组低(P<0. 05)o结论抗氧化剂Ebselen和VitE能降低NO缺乏性大鼠心肌组织中的氧化应激和 p22phox的表达,Ebselen作用优于VitE,两者联用作用优于单独使用。
关键词:一氧化氮;氧化应激;高血压;抗氧化剂;p22phoxinABSTRACTObjectTo observe antioxidant VitaminE and Ebselen on myocardium oxidative stress and the expression of p22phox in NO・ deficient Rats •Methods40 Wistar rats were divided randomly into 5 groups (n=8): Normal group(C); Experimental control group(L); Ebs group(S) Preventative ; VitE group(V) ; Ebs and VitE group (D), administered by gavage method. Their weight and systolic blood pressure (SBP) were measured fortnightly. The rats were killed after 8 weeks, homogenate of myocardium were collected for biochemical examination such as NO, Angll^MDA and O^. Measure the ratio of heart weight- body weight (HW/BW). The expression of p22phox protein in myocardium were measured by Western blotting.Results① SBP of the rats increased gradually .After 8 weeks, SBP of all rats in experimental groups was obviously higher compared with that of C (P<0.01), and SBP of the antioxidant teams was lower than that of L (P<0.05).. SBP of D was obviously lower than that of S (pvO.Ol), and SBP of S was fairly lower than that of V (PvO.05)•②The NO level, in the tissue of heart of each experimental group were obviously lower than those in C, and those in the antioxidant groups were predominantly higher than those in L(p<0.05), the combination groups more higher (p<0.05).And those in Ebselen groups were higher than those in VitE groups (p<0.05).The level of Ang II >MDA , O2" in the tissue of heart,the ratio of heart weight-body weight (HW/BW) of each experimental group were obviously increased than C (p<0.05), the combination administration group increased less than the single administration groups, and Ebselen groups increased less than VitE groups (p<0.05). the ratio of heart weight-body weight (HW/BW) of each experimental group were obviously increased than C (p<0.05), the combination administration group increased less than the single administration groups, and Ebselen groups increased less than VitE groups (p<0.05)/⑤the expression of p22phox of each experimental group were obviously increased than C (p<0.05), the combination administration group increased less than the single administration groups, and Ebselen groups increased less than VitE groups (p<・5)・ConclusionEbselen and VitE can decreas the cardiac oxidative stress and the expression of p22phox in NO- deficient Rats .Ebselen is better than VitE, and the combination is better than single used;Key Words: Nitric Oxide, oxidative stress, Hypertension, Antioxidant ,p22hox.学位论文独创性声明学位论文独创性声明本人声明所呈交的学位论文是本人在导师指导下进行的研究工作及取得的 研究成果。

动物 实验研究表明,当持续给予大鼠NO合酶抑制剂后,动物出现血压升高、血管周 围纤维化、心肌肥厚、心肌坏死及纤维化等心血管重构病变4】,是一种公认的研 究髙血压性心脏损害模型高血压、冠心病.心力衰竭等心血管疾病中普遍存在氧化应激(oxidative stress)o活性氧产生和清除的异常与高血压病、冠心病等血管损伤密切相关I) 研究证实,血管壁各种细胞中NADPH (还原型型辅酶II)氧化酶是活性氧 (reactive oxygen species ROS)产生的主要调控酶之一NADPH氧化酶包括5 个亚单位,即 gp91-phox、P22-phox、P47-phox、P67-phox、P40-phox ・ p22phox 是NADPH氧化酶的重要亚单位,在氧自由基生成中起关键性作用血管紧张素 II (Ang II)是NADH (还原型辅酶I) / NADPH (还原型辅酶II )氧化酶重要的刺 激因子在AT受体的作用下,Ang II可促进p22phox表达增强,使氧自由基产 生增加动物实验已经证实,抗氧化剂可以通过减轻氧化应激而起到减轻心脏损害 的作用维生素E是一种经典的抗氧化剂,它的抗氧化作用主要是在细胞 膜上,可清除细胞膜上的氧自由基,阻止膜或脂蛋白上的多不饱和脂肪酸发生氧 化反应。
以往的临床和动物实验表明,维生素E能够降低机体内活性氧生成,减 轻机体的氧化应激心⑷o Ebselen(Ebs)是一种具有拟谷胱甘肽过氧化物酶 (GSH-PX)活性的含硒化合物,也是一种经典的抗氧化剂,其抗氧化作用主要是在细胞液中,通过中和。
