    • 甘露消毒丹及其挥发油对流感病毒感染小鼠肺部炎症相关因子的影响600字    目的:?^察甘露消毒丹(Ganlu Xiaodu Mircropill,GXM)及其挥发油对流感病毒(influenza virus,IV)感染小鼠肺组织病理炎症、血清IL-6、TNF-α含量及肺组织NS1、NF-κB表达的影响,探讨其抗IV感染的机制方法:SPF级BALB/c小鼠108只,随机分为正常组(A组)、模型组(B组)、GXM灌胃组(C组)、GXM滴鼻组(D组)、GXM合用组(E组)、奥司他韦组(F组),除A组外,其余5组以IV FM1-6-E2滴鼻造模于造模后第3、7、10天进行取材,对小鼠肺组织病理炎症程度进行评分,并计算肺指数及干湿比,在电镜下观察肺组织超微结构,采用ELISA法检测血清中IL-6、TNF-α的含量,采用qPCR方法检测肺组织中NS1的基因表达,采用Western Blot法检测肺组织NF-κB的表达结果:IV感染后,小鼠肺组织出现炎症改变,其肺组织病理评分、肺指数均升高,干湿比降低,第7天炎症最为严重电镜下可见肺泡上皮细胞肿胀,纤毛减少、脱落,细胞器减少血清IL-6、TNF-α含量均升高,肺组织NF-κB表达升高,于第3天达峰值,而后降低;NS1表达亦升高,之后逐步下降。

      与B组比较,各治疗组均能够减轻肺部炎症,降低病理评分、肺指数,升高干湿比,降低TNF-α含量,下调NF-κB表达,其中尤以E、F组作用最为明显(P<0.05)治疗第3天,与B组比较,D、E、F组均能下调NS1的表达(P0.05);C、E、F组均能够降低IL-6含量(P0.05)治疗第7天,C组能够下调NS1表达,D组能够降低IL-6含量(P<0.05)结论:GXM可降低IV感染后肺部炎症,降低NS1蛋白的表达;减少促炎因子的释放,其滴鼻灌胃合用较单一给药有优势 毕业   流感病毒; 甘露消毒丹; 挥发油; 细胞因子  Effects of Ganlu Xiaodu Micropill and Its Volatile Oil on Lung Inflammation Related Factors in Mice Infected with Influenza Virus/LIU Guanghua,JIA Xiaoru,MIN Na,et al.//Medical Innovation of China,2018,15(03):001-006   Objective:To observe the effects of Ganlu Xiaodu Micropill(GXM) and its volatile oil on pathological inflammation,IL-6 and TNF-α in serum,and the expression of NS1 and NF-κB in lung tissue of mice infected with influenza virus(IV),to explore the mechanism of anti IV infection.Method:108 SPF BALB/c mice were randomly divided into normal group (group A),model group(group B),GXM lavage group(group C),GXM nasal drops group(group D),GXM group(group E),Oseltamivir group(group F);except group A,the other  5 groups were made models with IV FM1-6-E2 nasal.On the third,seventh,tenth day after modeling,pathological inflammation were scored,lung index and the ratio of dry and wet in mice lung tissue were assessed,and ultra structure of lung tissue was observed under electron microscope,ELISA method was used to detect the content of IL-6 and TNF-α in serum,the expression of NS1 gene in lung tissue was detected by qPCR method, and NF-κB by Western Blot method.Result:After IV infection,inflammation changes in the lung tissue of mice,the lung pathological score and lung index increased;the ratio of dry and wet decreased,on seventh day,inflammation was the most serious.Under electron microscope,alveolar epithelial cells of mice lung tissue swelled,the cilia decreased and shedding,organelles decreased.The content of IL-6 and TNF-α in serum and the expression of NF-κB in lung tissue increased,and all reached peak on the third day,then decreased,the expression of NS1 increased in the same time.Compared with group B,the treatment groups can reduce lung inflammation,reduce the pathological score and lung index,rise the ratio of dry and wet,reduce the content of TNF-α,cut the expression of NF-κB,especially in group E and F were the most obvious(P<0.05).On the third day of treatment,the expression of NS1 was down regulated in group D,E and F(P0.05).Group C, E and F could reduce the content of IL-6(P0.05).On the seventh day of treatment,group C could reduce the expression of NS1,and group D could reduce the content of IL-6(P<0.05).Conclusion:GXM can reduce lung inflammation and the expression of NS1 protein after IV infection,and reduce the release of proinflammatory factors,the combination of nasal drops and lavage administration has advantage over single administration.       临床表现轻重不一,轻者仅表现为感冒,病程短、症状轻、预后良好,重者可发展为流感病毒性肺炎,甚则危及生命[2]中医药在防治IV感染方面具有重要的作用,临床多以辛凉解表或清热解毒之品治之每获较好疗效但近年由于雾霾等环境污染及人们生活方式的改变、饮食的偏嗜,体虚湿盛或湿热内蕴者日益增多,IV等病原微生物极易附着于PM2.5等微尘颗粒侵袭人体,并因人的体质而从化,发为湿热性质的病证临床已有报道用甘露消毒丹(Ganlu Xiaodu Micropill,GXM)治疗IV感染取得了较好的疗效[3]本研究从湿热蕴毒立论,探讨清热解毒、利湿化浊的GXM滴鼻、灌胃两种不同给药途径对IV感染小鼠肺部炎症的影响通过观察小鼠肺组织病理炎症、肺指数、干湿比、肺组织超微结构、血清中IL-6、TNF-α含量变化及肺组织中NS1、NF-κB表达,揭示GXM一方两用对IV感染的效用机制  1 材料与方法  1.1 材料  1.1.1 病毒株 甲型流感病毒鼠肺适应株A/FM1/47(H1N1),由辽宁中医药大学附属医院病毒室保存  1.1.2 实验动物 SPF级BALB/c小鼠108只,体重(20±2)g,雌雄各半,购买于辽宁长生生物技术有限公司,生产合格证号:SCXK(辽)2013-009,使用合格证号:SYXK(辽)2015-0001。

        1.1.3 药物及制备 GXM药物组成:滑石、黄芩、茵陈、石菖蒲、川贝母、木通、藿香、连翘、白蔻仁、薄荷、射干,中药饮片购于辽宁中医药大学附属医院门诊药局,经辽宁中医药大学中药分析教研室鉴定为正品饮片以8倍量70%乙醇加热回流提取3次,1 h/次,合并每组3次提取的药液,回收溶剂并浓缩,制成含生药1 g/mL混悬液挥发油制备:将中药粉碎,加8倍量水浸泡,装入冷凝器中,水蒸气蒸馏提取6 h,分层分取挥?l油,制成含生药6 g/mL的药液磷酸奥司他韦颗粒(生产厂家:宜昌长江药业,批准文号:国药准字H2009372)75 mg,配成1 mg/mL的溶液每组药液均4 ℃保存备用  1.1.4 试剂与仪器 Trizol总RNA提取试剂,Invitrogen公司;RT-PCR试剂盒、DEPC、DNAmarker DL2000,购于大连宝生物公司;小鼠IL-6、TNF-α ELISA试剂盒,Phospho-NF-κB p65抗体试剂盒,GAPDH,ECMII of Western-blot检测试剂盒浓缩型免IgG二抗,购于武汉博士德公司  1.1.5 主要仪器 TP1020型自动组织脱水处理机;EG1150型石蜡包埋机(LEICA,德国);RM2135型病理精密轮转切片机(LEICA,德国);BX50F4型显微镜(OLY-MPUS);荧光定量PCR仪(LC-Ⅱ);IM71型酶标仪;PowerPac Universal电泳通用型电源;Trans-Blot SD半干转印系统(Bio-Rad,美国);FlourChem Q蛋白印记成像系统(Proteinsimple,美国);低温高速离心机(Heraeus,德国);H-7650型透射电镜(日本株式会社)等。

        1.2 实验方法  1.2.1 动物分组 用随机数字表法将小鼠。
